Abstract

Background:Currently,high-densitylipoproteincholesterol(HDL-C),afactorwhichpreventsprogressionofarteriosclerosis,ismeasuredusinglaboratory-basedchemistryanalyzerswithoutapretreatmentstep.BecauseHDL-Cismeasuredwithapretreatmentstepinmanypoint-of-caretestingsystems,adirectassayisneeded.Methods:Adry-chemistry-basedassayusingsurfactantshasrecentlybeendevelopedinparallelwiththedevelopmentofadedicatedreagent.Asimpleanalyzerthatacceptswholebloodsampleswasalsodeveloped.Results:Theassaydemonstratedexcellentprecision,dilutionlinearityandintermethodcomparison.Inaninterferencetest,assayvaluestendedtobelowerinthepresenceofhighconcentrationsofhemoglobin,conjugatedorunconjugatedbilirubin.Neitherascorbicacidupto20mg/dl,norformazinturbidityupto2100,hadaneffectontheassay.Conclusions:Thisdry-chemistryassayusingonlysurfactantsforspecificityinthedirectHDL-Cmethodwasjudgedusefulforpoint-of-careinstrumentationintermsofequipmentcompactness,operationalsimplicityandrapidresponsiveness.D2002ElsevierScienceB.V.Allrightsreserved.
